A brand new research from The Hospital for Sick Youngsters (SickKids) and Institut Curie reveals how stem cells sense and reply to their surroundings, with implications for inflammatory bowel illness and colorectal most cancers.

Stem cells continuously adapt to their surroundings to keep up organ and tissue well being, knowledgeable by chemical alerts and bodily forces. When they don’t perform as meant, stem cells may end up in quite a lot of well being situations together with inflammatory bowel illness (IBD) and colorectal (bowel) most cancers, the place they proceed to divide till a tumor varieties.

Till now, how stem cells sense the bodily forces round them has remained unclear, however novel findings printed in Science led by Dr. Meryem Baghdadi, a former SickKids postdoctoral researcher, Dr. Tae-Hee Kim at SickKids and Dr. Danijela Vignjevic at Institut Curie, has revealed that stem cells rely on two ion channels, referred to as PIEZO1 and PIEZO2, for his or her survival.

“The physical properties of the environment around stem cells are crucial to our health,” explains Kim, Senior Scientist within the Developmental & Stem Cell Biology program. “With this knowledge, we can explore ways to promote gastrointestinal regeneration to not only prevent, but repair damaged stem cells.”

How stem cells sense modifications in our intestine

In 2018, Dr. Xi Huang, Senior Scientist within the Developmental & Stem Cell Biology program at SickKids, discovered that PIEZO ion channels affect tumor stiffening in mind most cancers. Impressed by this analysis, Kim’s analysis group got down to discover how stem cells within the intestines use PIEZO channels to remain wholesome and performance correctly.

In a preclinical mannequin, the research group knocked out (turned off) PIEZO1 and PIEZO2 within the intestines. The outcomes have been dramatic: within the absence of each PIEZO channels, the stem cells could not keep their essential features, resulting in extreme sickness and fast demise. Though these PIEZO channels have been beforehand recognized to have distinct features, this research has revealed their surprising redundancy in stem cell upkeep.

The Kim and Vignjevic labs recognized that PIEZO ion channels have been serving to stem cells really feel bodily modifications of their environment, like how stiff or stretchy the surroundings is. With out these channels, there was an imbalance in two crucial signaling pathways, inflicting the stem cells to overlook necessary modifications of their surroundings and improperly differentiate.

“When PIEZO channels are missing, stem cells can’t stay stem cells. Instead, they turn into other cell types too quickly, leading to serious health problems,” says Baghdadi.

The invention has important implications for human well being, significantly for situations with irregular stem cell exercise similar to IBD, one of many quickest rising situations in Canada, and bowel most cancers, the third most typical most cancers within the nation.

“More and more we are finding that our cells are more than just biology, there are chemical and mechanical signals which are driving cell activity,” says Kim. “How and why our body responds to these signals will open up new doors of research not just for gut health, but for every aspect of human health.”
